Aral Balkan - A happy grain of sand: http://www.besquare.me/session/a-happy-grain-of-sand/ He did this talk at various conferences in various variations. Some extremely good thoughts in how to use our skills to make better products and thus making lives better for everyone. Aral is probably one of the most inspiring people I ever had the pleasure to listen to, and partly thanks to him I sure as hell wasn't on the path I am on today.
Bret Victor - Inventing on Principle: http://vimeo.com/36579366 Powerful talk on designing and developing and essentially living your life based on guiding principles. Still gives me goosebumps.
Joe Stewart @ Creative Mornings Los Angeles: http://vimeo.com/58672952 A good designer friend of mine suggested this one. Incredible good talk on design and inspiration and not giving up when it seems people don't give a damn about your work. Note: some profanity ahead.

Surprised nobody has mentioned the Aaron Draplin Creative Mornings talk. He has a really unique perspective and way of communicating, both sorta folksy-funny and poignant at the same time.

Most of the good product/UI/UX design talks that came to my mind have already been mentioned but this one is a classic that I used to watch back in my design school days when I wanted to be a graphic designer. I think the bigger picture is still applicable to product design and it's just a joy to watch. If Wilson Miner's talk moves me to tears this one without fail always makes me smile.
I always ask people who design to watch Scott Berkun (ex Microsoft IE lead but don't hold that against him) talk about the Myth of innovation. I have met too many designers feeling like they haven't had "the lightbulb" moment and he talks about how it doesn't exist.
